Bike not easily turning over after ride

I recently built a 100cc motorized bike, after completion it ran and rode good. After one of the first short rides I shut off the engine. I go to start it again and it locks up the back tire and barely turns over. The engine turns freely in reverse and with the spark plug out. It acts like the piston is locked up but it’s not and the cylinder walls aren’t scored or pitted since it doesn’t even have a hour of riding time on it. It seems like it’s got too much compression and it just locks up the rear tire even though it was starting/running just fine when I shut it off. It hasn’t been on any long trips, only short ones around the yard or down the road, I know it’s not the gas mix ratio since I put about 6.5 ounces of 2 stroke oil to 1 gallon of gas.
I am quite knowledgeable in anything to do with engines I just can’t figure out what’s going on. Please help!
