What are the best parts you have found to increase the comfort of riding? Best seat, grips, reduced vibration, sound or anything else

My home made saddles. I use what the upholstery guys call 'motorcycle foam' which is a very high density foam rubber. 2" is nice but 3" feels like sitting on a cloud.

Many won't agree but I really like the ride a set of ape hangers gives. They increase my riding comfort on long straight rides. They just feel right.

Big fat soft tires are also a real plus when it comes to making for a comfy ride.
And, room to stretch out my legs.

Yup... fat tires, front suspension, mid-size apes, and a decent seat.

Low bars set you up for a launch-on-impact scenario!
BTR may be super-cool, but just not practical off the track.

my mini ape hangers on my ambassador are really comfortable ....then the big cushy seat

Just to add, a layback seatpost is an awesome accoutrement. Can get really tiring holding your self up on your hands and feet when you are almost over the pedals. Makes a huge difference to be actually sitting rather then more propped up.

I had no idea what a difference one would make until Tom sent me one he made.

if I had to pick the most important comfort item, it would be a suspension fork. All the others items mentioned helped me too, but not as much.
